A 2026 study characterized MOTS-c as an amphipathic, cationic peptide with direct antibacterial activity. In vitro, MOTS-c targeted Escherichia coli and methicillin-resistant Staphylococcus aureus (MRSA) membranes via its hydrophobic and cationic domains. In a murine acute peritonitis model, MOTS-c neutralized MRSA infectivity. In human monocyte cultures, interferon-γ, lipopolysaccharides, and differentiation signals each induced endogenous MOTS-c expression. Exogenous MOTS-c applied during primary mouse monocyte differentiation reprogrammed cells into macrophages with distinct transcriptomic signatures related to antigen presentation and interferon signaling, with shifted metabolic profiles and altered bacterial clearance capacity [1].
