ARA-290 16MG provides ARA-290 (Cibinetide) as lyophilized powder in a 3 mL vial.

Receptor Selectivity and Signaling Context

ARA-290 is studied in nonclinical systems as a ligand for the EPOR/CD131 heteromeric receptor complex. Downstream signaling cascades investigated in experimental models include JAK2, PI3K/Akt, and NF-κB-associated pathways. This reported receptor selectivity distinguishes ARA-290 from full-length EPO in experimental paradigms examining non-erythropoietic receptor-mediated signaling.

Selected Nonclinical Research Findings

In Vitro: Cisplatin-Induced Cell Stress Model

Ghassemi-Barghi et al. (2023) evaluated ARA-290 in HEK-293 and ACHN cell lines exposed to cisplatin. ARA-290 pretreatment was associated with reduced DNA damage parameters (comet assay, micronucleus frequency), decreased malondialdehyde and reactive oxygen species levels, and increased antioxidant enzyme activity compared to cisplatin-only controls. Pro-inflammatory cytokine levels (TNFα, IL-6, IL-1β) and caspase-3 activity were lower in ARA-290-treated groups. Gene and protein expression analysis showed decreased Bax and increased Bcl2 levels relative to cisplatin-only groups [36085231].

In Vitro and In Vivo: Osteoclastogenesis and Bone Measurements

Awida et al. (2021) examined cibinetide in murine bone marrow-derived osteoclast precursor cultures and in C57BL/6J female mice. In vitro, cibinetide inhibited osteoclastogenesis when administered alone or in combination with EPO. In vivo, cibinetide-treated mice showed higher cortical and trabecular bone mineral density measurements compared to controls. Flow cytometry indicated reduced osteoclast progenitor populations (Lin⁻ CD11b⁻ Ly6Cʰⁱ CD115⁺) in mice receiving cibinetide plus EPO versus EPO alone [35008482].

In Vivo: Monocyte Modulation in APP/PS1 Mice

Al-Onaizi et al. (2022) administered ARA-290 systemically to young APP/PS1 transgenic mice. ARA-290 increased circulating Ly6Cˡᵒʷ patrolling monocyte levels and monocyte progenitors in bone marrow. Amyloid-β burden was reduced in treated animals compared to controls. In chimeric APP/PS1 mice with selective Ly6Cˡᵒʷ monocyte depletion, ARA-290 did not produce these changes, suggesting dependence on this monocyte subset. ARA-290 was not associated with similar outcomes when administered to aged APP/PS1 mice with advanced pathology [34343617].
